Standing at 1.93 meters (6 feet 4 inches) tall and weighing 103 kilograms (227 pounds), he plays as an opposite spiker. Darlan began his volleyball journey in 2017 with the youth team of Fluminense FC. In 2018, he transferred to Sesi-Bauru, where he was promoted to the senior team in 2019. He is the younger brother of Alan Souza, who also plays as an opposite hitter for the Brazilian national team.

In 2018, Darlan led the Brazilian U19 national team to victory at the South American Championship and was named the tournament's Most Valuable Player. He participated in the 2019 FIVB Volleyball Boys' U19 World Championship, where Brazil finished ninth. In 2021, he competed in the FIVB Volleyball Men's U21 World Championship, helping Brazil secure a seventh-place finish.

Darlan made his debut with the Brazilian senior national team in 2022 during the Volleyball Nations League (VNL). That same year, he played a crucial role in Brazil's bronze medal win at the FIVB Volleyball Men's World Championship. In 2023, he was instrumental in Brazil's qualification for the Paris 2024 Olympics, emerging as the team's top scorer during the Olympic Qualifying Tournament.

During the 2023 Pan American Games, Darlan helped Brazil clinch the gold medal and was honored with the Best Opposite and Best Server awards. In the 2023-2024 Superliga season, he set a new league record by scoring 41 points in a single match, surpassing the previous record of 40 points. He later broke his own record by scoring 48 points in a semifinal match. Darlan's exceptional performance led Sesi-Bauru to the Superliga championship, and he was named both the Most Valuable Player and Best Opposite of the season.

In early 2025, reports indicated that Darlan would join the Italian club Verona Volley for the upcoming season.
